The focus of the Saper laboratory is on the brain circuitry that regulates key functions including regulation of wake-sleep cycles, circadian rhythms, body temperature, and feeding. We use a variety of methods to analyze neuronal circuits and their functions, including viral vectors and transgenic mice to manipulate neurons opto- or chemogenetically, trace their connections, and record from neurons using calcium imaging. In addition to identifying this circuitry in experimental animals, we also are interested in determining the homologous circuitry in human brains, and in determining how it may be disrupted in specific neurological and psychiatric disorders.
