Narrative Description of Research:

My laboratory currently has active projects in three areas: mitochondrial roles in neurologic injury, metabolomics and proteomics for personalized risk analysis, and the analysis of megavariate data sets.

There are no drugs currently available that protect the brains of people who have strokes, head injuries or neurodegenerative diseases, such as Alzheimer’s or Parkinson’s from further degeneration. We have identified a series of mitochondrially targeted, FDA-approved drugs that are being tested in a series of biochemical, cell and animal models. Initially we have examined over 1,000 different drugs and identified those that affect specific subcellular events believed to play roles in cell death caused by stroke, head injury, and neurodegeneration. These studies identified a series of drugs that delay the onset of neurodegeneration in both cell and animal models of stroke. Our data indicates that these drugs may lessen post-stroke damage and delay disease progression in humans. While other effects of these drugs must be considered, the fact that many of these agents are already in use suggests the potential to move these agents from “bench to bedside” with relative quickness. We have further developed analogs of these agents that may be more effective.

My laboratory is also testing whether blood tests based on studies of long-term food intake in rats can enable scientists to predict risk of human disease. Rats maintained on long-term, low calorie diets have reduced risk of certain diseases, such as cancer. This is analogous to the reduced risk of diseases seen in humans who are comparatively lean. My collaborators and I have developed approaches to simultaneously examine over 1,000 molecules in the blood. We then use mathematical approaches to identify sets of these compounds that can be used as dietary biomarkers (giving information about the diet). Our goal is to be able to use blood tests developed in these studies to predict long-term disease risk in individuals and use this information for possible protective interventions and increased monitoring. We are initially focused on diseases where preventative intervention and/or early diagnostic screening are available (eg, breast cancer, Type II diabetes).

We have also developed expertise in high data density informatics to support the medium- throughput studies we carry out on mitochondria and the high data density metabolomics studies that we are pursuing. As this side of our research has progressed, we are now turning the tools that we developed to address issues in clinical informatics, for example we are using these tools to understand the factors that modulate recovery (rehabilitation) following traumatic brain injury.

I am the Deputy Director of Harvard Gene Therapy Initiative, an organization founded in 1998 within Harvard Medical School with the objective of promoting the use of gene transfer technology in both research and therapeutic applications. Over the last nine years, my group has been primarily responsible for the development of three cell based products intended for cancer immunotherapy, the development of both research and clinical grade production of adeno-associated viral vectors targeting neuromuscular systems, as well as newer generation lentiviral vector systems. The work has supported two approved investigational new drug (IND) applications in the United States and Europe, and several more in progress, in collaboration with physician scientists at Dana Farber Cancer Institute and elsewhere.

Before joining Harvard Gene Therapy Initiative in 1999, I received post-doctoral training at Whitehead Institute, MIT, and Children’s Hospital, Boston. Prior to receiving my PhD in Virology in 1995, I received my MD from National Taiwan University in 1990.

----My laboratory studies how DNA damage response genes maintain genome stability to suppress cancer and other age-associated disorders. Previous research has shown that the ATR protein kinase mediates a signaling pathway that prevents double strand break formation in the event that DNA replication stalls. Since DNA replication stalling occurs in every cell cycle at sites of normal oxidative DNA damage, failure of the ATR-mediated pathway to function properly at these sites may be a major source of genomic instability that causes cancer and age-related diseases.

----Generated two mouse models that disrupt the ATR checkpoint kinase and studied the effects of deletion on cell cycle checkpoints, checkpoint signaling, genome stability, embryonic development, and tumorigensis. ATR is required for genome maintenance and tumor suppression. Cre/lox conditional deletion of ATR leads to loss of cell cycle checkpoints and genome stability during S phase.

I did a Post Doc at the Dana Farber Cancer Center with John Gribben, MD, PhD. Here I developed and improved methods for detecting residual disease in leukemia and lymphoma as well as investigated the potential use of data from these assays in making treatment decisions for patients undergoing chemotherapy.

I joined Millennium in 2000 as a Scientist in their Process Technology group where I provided assay development, automation and process improvement support for the Discovery organization. I am now a member of their Lead Discovery group where I head the automation group. We provide automation support for high throughput screening and other assays that support early drug discovery. In addition to this role I also interact with Scientist across the company to help develop automation friendly assays.

I continued with the same firm after graduation and became a partner in the firm. My current legal practice involves primarily patent litigation and prelitigation counseling as well as intellectual property due diligence investigations in the area of pharmaceuticals, biologics, and life science diagnostics.

After graduating from the DMS Virology program, I worked for two years with the Department of Cell Biology at Harvard Medical School as their scientific IT systems manager, working with labs to more effectively utilize IT in their research. In 1997, I was appointed Director of the Research Computing Center, an HMS-wide department which provided research application support, training, and in-house consultation. In 2002, I left HMS to join a technology consulting company, and worked at Biogen Idec, Inc. of Cambridge, MA. During this time I started a small company to provide technology integration and computational biology research services. In 2004, I rejoined HMS as a part-time scientist.

I am currently a staff scientist in Mark Kircshner's lab in the Department of Systems Biology at Harvard Medical School, part of a multi-geographic team working on the hemichordate Saccoglossus kowalevskii (common name, the Acorn worm). As a means to understanding the evolution of chordates and the early bilateral ancestor, we are studying the expression of key genes and developmental circuits in early development through EST sequencing, in situs, and microarrays.

The usual lab work schedule is pleasantly interrupted each year in September, at which time we relocate to the Marine Biology Labs in Woods Hole, MA to work with live animals. Designated a model organism by the NIH, the genome is currently being sequenced and we hope to have the full assembly by the end of 2007. My responsibilities are a) analyzing and managing all the sequence and expression data for the project team, b) to design and deploy visualization tools, c) lead person for the microarray design and analysis, and d) lead team member for the logistics for the genome annotation.

After receiving my degree, I finished work in my thesis lab. Then I joined Hazel Sive's laboratory at the Whitehead for a postdoc position since I was very interested in getting back into developmental biology. My research focused on the molecular basis of early neural development in the frog Xenopus. My husband and I were out of phase so that I became a trailing spouse, leaving my postdoc position prematurely to move to Bozeman and begin a research track position at Montana State University (I also had my three children during this time). I was able to obtain a tenure track position at Montana State University in 2002. The work in my laboratory focuses on the molecular networks that govern early neural development in Xenopus and chick embyros.
